Cases in the capital have soared in recent weeks forcing London, along with parts of the South-east, to be pushed into Tier 3 restrictions.
Ahead of Christmas next week, Mr Khan warned the public to be extra careful as the NHS struggles to cope with the virus.
He said: “Our NHS is second to none in the world, but rising COVID cases are pushing it to its limits.
“We must bring our cases down urgently, otherwise we risk overwhelming our NHS and losing more lives.
“This can’t happen. Please, do not take any unnecessary risks this weekend.”
Cases have skyrocketed in the capital, with City Hall reporting 29,144 people tested positive in London between December 6-12.
